Diego was a royal ambassador, lawyer and Lord of Agoncillo, Velilla and San Martín de Berberana in the Kingdom of Castile and León. He is not to be confused with his nephew, Don Diego López de Medrano y Zúñiga, Lord of Fuenmayor and Almarza de Cameros, son of his brother Don Juan Martínez de Medrano y Hurtado
